Purification and biochemical characterization of polygalacturonase produced by penicillium expansum during postharvest decay of 'Anjou' pear.
A polygalacturonase (PG) was extracted and purified from decayed tissue of 'Anjou' pear fruit inoculated with Penicillium expansum. Ammonium sulfate precipitation, gel filtration, and cation exchange chromatography were used to purify the enzyme. Both chromatographic methods revealed a single peak corresponding to PG activity. PG enzyme activity from healthy and wounded pear tissue was undetect...
متن کاملFumigation of Fruit with Acetic Acid to Prevent Postharvest Decay
Acetic acid (AA) as a vapor at low concentrations was effective in preventing fruit decay by postharvest fungi. Fumigation with 2.7 or 5.4 mg AA/liter in air at 2 and 20C reduced germination of Botrytis cinerea Pers. and Penicillium expansum Link conidia to zero after they had been dried on 0.5-cm square pieces of dialysis tubing. Decay of ‘Golden Delicious’, ‘Red Delicious’, and ‘Spartan’ appl...
متن کاملIsolation and Characterization of Polygalacturonase Produced by Tetracoccosporium sp.
Thirty-five fungal strains which isolated from vegetable wastes, were screened for the use of polygalacturonic acid as the sole carbon source. Twenty-five isolates were positive for polygalacturonase activity in cup-plate assay, as evidenced by clear hydrolysation zones. The most productive strain was determined by measuring clear zones formed around colonies stained with ruthenium red.</em...
متن کاملThe purification, physical/chemical characterization, and cDNA sequence of cantaloupe fruit polygalacturonase-inhibiting protein.
ABSTRACT Polygalacturonase-inhibiting proteins (PGIPs) are believed to aid in plant defense against fungal pathogens by inhibiting polygalacturonases (PGs) secreted by the invading organism. In an effort to better understand this type of plant-pathogen interaction in cucurbits, we have isolated a cantaloupe PGIP (CmPGIP) from 5 to 15 day postanthesis cantaloupe fruit. CmPGIP inhibited crude ext...
متن کاملControl of postharvest decay of citrus fruit with calcium polysulfide
Incidence of green mold of citrus, caused by Penicillium digitatum, was reduced by 80% or more by the immersion of lemons or oranges for 1–4 min in warm (40.6–43.3°C) ‘liquid lime-sulfur’ (LLS) solution that contained 0.75% (wt vol) calcium polysulfide. The incidence of sour rot, caused by Geotrichum citri-aurantii, was reduced 35–70% by this treatment. LLS was similar in effectiveness to other...
